TITLE: PREPARATION OF CELL BANKS FOR DISTRIBUTION STOCKS


INTRODUCTION

The purpose of this protocol is to describe the procedures to be followed when preparing cell banks for distribution.

PROCEDURE

For new lines into the Collection or if existing stocks have reached less than 10 ampoules in distribution bank, remove one vial from a fully Quality Controlled Master stock.

Resuscitate (according to
AHC/1998/3/2.5/1, AHC/1998/3/2.5/2) and grow cell line with reference to safety data sheet and protocol AHC/1998/3/2.1/2 on routine sub-culture of cell lines.

Grow cell line to produce approximately 20 to 50 ampoules at between 2 to 4 x 106 cells per ampoule at 80 to 100% viability (cell counting method AHC/1998/3/3.1/2.1).

Prepare labels with cell line name, CB number, passage number if known, and date.

Cryopreserve cells according to
AHC/1998/3/2.4/1 and AHC/1998/3/2.4/2.

Store ampoules in the appropriate location in the liquid nitrogen tanks and record locations.

Update the relevant databases, giving batch number, location, batch of source material, number of ampoules and passage number if known.
