LABORATORY PROCEDURES FOR PLANT CELL LINES

REFERENCE NO.: PC/1998/3/4.1

TITLE: DESCRIPTION AND USE OF THE COMPUTER DATABASE FOR CRYOCONTAINER MANAGEMENT

INTRODUCTION

After cryopreservation the frozen ampoules for the "Master-" and "Working Collection" are stored in cryocontainers. To localise the position, where ampoules of a specific cell line are stored in the cryocontainer a computer data base is used.

The storage system consists of two cryocontainers, each containing 6 towers, each containing 8 trays, each containing positions for 78 samples. Each tray is described by four pages in the computer data base. The first page contains a diagram indicating a code number for the position of each vial and a list for the entry of the short name or abbreviation for each vial. The next three pages contain a list to enter a full name for each vial. The name of the data base is cryo.con. It has the format of a WordPerfect 6.1 file.

PROCEDURE

1. Load the file cryo.con (path: f:\user\plant\cryo.con)

2. Move the cursor to the page where you want to enter data.

3. Enter the new data into the file cryo.con:

3.1. Enter a short name for the samples into the list behind the number for the corresponding position.

3.2. Enter the full name of samples on the next pages for the specific tray behind the number for the corresponding position.

4. If samples/ampoules are taken out of a cryocontainer data have to be deleted as follows:

4.1. Delete the short name for the samples that have been taken out from the list behind the number for the corresponding position.

4.2. Delete the full name of the samples that have been taken out on the next pages for the specific tray behind the number for the corresponding position.

5. If all the names for all samples in all their positions have been entered resp. deleted, store the modified data base file under the name cryo.con (path: f:\user \plant\cryo.con).

6. Make a printout of the 4 pages for those trays where data have been changed and mark it with the present date.

7. Close the file cryo.con.
